In a support mission, you will usually be riding atop a vehicle while shooting away at zombies attacking your subject. This will be your first support mission, wherein you will be protecting someone (either a comrade or a survivor). Note: After this mission, you will successfully be promoted to Rank 2 and you will also receive a special weapon (an M240) which will be needed for the next mission.
